Lipid metabolism of rumen ciliates and bacteria. I. Uptake of fatty acids by Isotricha prostoma and Entodinium simplex.

چکیده
Washed suspensions of the ruminal ciliates, Isotricha prostoma and Entodinium simplex, concentrated C(14)-labeled oleic, palmitic, stearic, and linoleic acids within the cells during short incubation periods. Radioautographs demonstrated that oleic acid-1-C(14) was hydrogenated to stearic acid by I. prostoma, and Warburg manometric data showed that the sodium salts of oleic, valeric, caproic, and acetic acids, and methyl myristate, methyl laurate, and the triglyceride tributyrin stimulated fermentation of I. prostoma. The total lipid and free fatty acid contents of I. prostoma were determined.

متن کاملRole of the protozoan Isotricha prostoma, liquid-, and solid-associated bacteria in rumen biohydrogenation of linoleic acid.
From the simultaneous accumulation of hydrogenation intermediates and the disappearance of Isotricha prostoma after algae supplementation, we suggested a role of this ciliate and/or its associated bacteria in rumen biohydrogenation of unsaturated fatty acids. The experiments described here evaluated the role of I. prostoma and/or its associated endogenous and exogenous bacteria in rumen biohydr...
متن کاملRumen Bacterial and Protozoal Responses to Insecticide Substrates.
Insecticides containing organophosphate, chlorinated hydrocarbon, and carbamate were tested with bovine ruminal ingesta fractions. Rumen bacteria exposed to insecticide levels of 0 to 500 ppm in rumen fluid for 4 hr were inoculated into rumen fluid-starch feed extract medium. No apparent significant bacterial count inhibitions were noted. Also, when insecticides were used as carbon sources at c...
متن کاملDigestion of a dehydrated lucerne/barley diet (70:30) in defaunated, Isotricha-monoinoculated and mixed-fauna-inoculated rumen in sheep.
Two adult Texel sheep fitted with permanent rumen fistulae were defaunated for 12 weeks in a preliminary experiment. They were then reinoculated with Isotricha sp for 11 weeks, and finally with a mixed fauna (Entodinium, Epidinium, Eudiplodinium and Isotricha) for 10 weeks. They were fed a diet composed of dehydrated lucerne (700 g/d), pelleted barley grain (300 g/d), grass hay (100 g/d) and wh...
متن کاملIntracellular Prokaryotes in Rumen Ciliate Protozoa: Detection by Confocal Laser Scanning Microscopy
In situ hybridization of rumen ciliate protozoa with 165 ribosomal RNA fluorescent oligonucleotide probes specific for Archaea and Bacteria provided semi-quantitative indication of the location, type and extent of prokaryotic colonization of various protozoal species. The isotrichid holotrich ciliates generally carried a smaller load of intracellular microorganisms than did the entodiniomorphid...
